Expanded Optogenetics Line

Nine headstages are now available for optogenetic studies in either tethered mice or rats. Most combinations of sleep, seizure, biosensor and optical stimulation can now be easily studied. Seven different wavelength LED probes are available which are compatible with most opsins.

To show you an example of the power of the system, we used a kindling model as part of our validation studies to generate seizures in a transgenic mouse [( Thy1-COP4/EYFP)9Gfng] expressing channelrhodopsin from Jackson Laboratory. The figure below shows a resultant seizure along with changes in glutamate.

Cortical EEG and hippocampal depth electrode activity was monitored along with prefrontal cortex glutamate changes. A 445 nm blue LED fiber probe was used for stimulation with light pulses at 20 Hz, 10% duty cycle, 600 mA, delivered for 20 seconds. Six stimulations occurred per day (30 minutes apart over a three-hour period). Initial LED power was 130 mW/mm².
